Stimulating Action of Angiotensin II on Calcium Channels in Vascular Smooth Muscle Cells

چکیده
The possible involvement of a GTP-binding protein in the regulation of Ca2' channels by angiotensin II (Ang II) in vascular muscle cells was investigated by the whole-cell voltage-clamp method. Single cells were freshly isolated from guinea pig portal vein. The pipette solution contained high Cs4 to inhibit K4 currents and thereby isolate the Ca2+ channel current. Ba24 (2 mM) was in the bath solution as a charge carrier for the Ca24 channel. Application of Ang

The effect of adrenomedullin and proadrenomedullin N- terminal 20 peptide on angiotensin II induced vascular smooth muscle cell proliferation
Objective(s): The study aimed to investigate the effects of adrenomedullin (ADM) and proadrenomedullin N- terminal 20 peptide (PAMP) on angiotensin II (AngII)-stimulated proliferation in vascular smooth muscle cells (VSMCs). Materials and Methods: Thoracic aorta was obtained from Wistar rats and VSMCs were isolated from aorta tissues and then cultured. In vitro cultured VSMCs were stimulated w...

The effects of 8-bromo-cGMP on intracellular calcium concentrations in cultured rat aortic smooth muscle cells were studied. Both angiotensin II and depolarizing concentrations of K+ stimulated Ca2+ accumulation in the cytoplasm. The increase in Ca2+ due to angiotensin II was associated with an increase in inositol phosphates, while that due to K+ was not. Preincubation of cells with 8-bromo-cG...
متن کاملAngiotensin increases cytosolic free calcium in cultured vascular smooth muscle cells.
We used the calcium-sensitive fluorescent indicator quin 2 to monitor changes in cytosolic free calcium concentration ([Ca2+]i) associated with angiotensin II receptor activation in cultured vascular smooth muscle cells isolated from rat aorta. Resting [Ca2+]i in unstimulated vascular smooth muscle cells was 198 +/- 7 nM. Angiotensin II induced concentration-dependent rapid increases in [Ca2+]i...
متن کاملIntracellular Angiotensin II Stimulates Voltage-Operated Ca Channels in Arterial Myocytes
Although the presence of intracellular angiotensin II (Ang II) and of Ang II–binding sites has been reported, their roles in cell function have not been fully clarified. The purpose of the present study was to test the hypothesis that intracellular Ang II modifies voltage-operated Ca channels in vascular smooth muscle. Ca channel currents were recorded in guinea pig mesenteric arterial myocytes...
متن کاملEffect of azelnidipine on angiotensin II-mediated growth-promoting signaling in vascular smooth muscle cells.
The detailed mechanism of the effects of extracellular Ca2+ entry blockade on angiotensin II (Ang II) type 1 (AT1) receptor-mediated growth-promoting signals in vascular smooth muscle cells (VSMCs) is not fully understood. Ang II stimulation caused biphasic activation of growth-promoting signals, reaching a peak at 5 to 10 min followed by a decrease and a second peak at around 2 to 4 h. Additio...
